Ipamorelin, the First Selective Growth Hormone Secretagogue

This study characterized ipamorelin as a selective growth hormone-releasing peptide using in vitro and in vivo experimental approaches. In vitro experiments using primary rat pituitary cells demonstrated that ipamorelin released growth hormone (GH) with potency and efficacy similar to GHRP-6 (EC50 = 1.3±0.4 nmol/L and Emax = 85±5% vs 2.2±0.3 nmol/L and 100%).

Pharmacological profiling using GHRP and growth hormone-releasing hormone (GHRH) antagonists demonstrated that ipamorelin stimulates GH release via a GHRP-like receptor. Specificity studies in swine showed that ipamorelin did not affect FSH, LH, PRL, or TSH plasma levels.

Unlike GHRP-6 and GHRP-2, which increased plasma ACTH and cortisol levels, ipamorelin did not release ACTH or cortisol at levels significantly different from those observed following GHRH stimulation. This selectivity was evident even at doses more than 200-fold higher than the ED50 for GH release. The researchers concluded that ipamorelin is the first GHRP-receptor agonist with GH release selectivity similar to that displayed by GHRH.
